Blow By Blow, winner of the Martin Pipe Conditional Jockeys' Hurdle at the Cheltenham Festival in March, will be bidding to make it two from two over fences in a race named in memory of former jockey and trainer Liam Brennan and his wife Valerie.
The Gordon Elliott-trained seven-year-old scored by 17 lengths on his chasing debut over the same trip at Galway last month and his trainer said: "Blow By Blow won well at Galway and has been in good form since. He won't mind the good ground.
"We also run Cubomania, who is four from six over fences this season. All those wins for us were in Britain and he has yet to win a chase here. He's in good order and likes good ground."
Orion D'Aubrelle, who got off the mark over fences at the Listowel festival in September, and Some Neck, who will be making his first appearance since May, represent Willie Mullins.
The champion trainer said: "Orion D'Aubrelle did the job nicely at Listowel. He'll like the trip and the ground should be fine for him.
"Based strictly on official ratings Some Neck would appear to have a good chance, but whether he is really a 140 horse is debatable. He probably needs slower ground."
Nick Lost, the mount of Rachael Blackmore and trained by Henry de Bromhead, returns from a two-month break and is two from three over fences. The six-year-old achieved his wins at Punchestown and Limerick during the summer.
Completing the six-horse line-up is Cap D'Aubois, a winner over this trip at Killarney in August and having his second run for Andy McNamara.
