Last year's winner will be part of Sir Henry Cecil's three-strong raid on the Group 1 contest, which will also include Frankel's pacemaker Bullet Train.
Twice Over saw off stablemate Midday to record one of the biggest victories of his admirable career under Ian Mongan in the race last season and Teddy Grimthorpe, racing manager to owner Khalid Abdullah, said: "Twice Over is the defending champion so it makes sense for him to run."
Frankel took his brilliant unbeaten record to 12 when securing back-to-back wins in the Qipco Sussex Stakes at Glorious Goodwood earlier this month and he will have his first start beyond a mile when he tackles the 1m2½f trip next week.
Grimthorpe added: "I don't think the step up to ten furlongs will be a concern. In his early days he used to get carried away with his phenomenal stride but under Sir Henry Cecil's guidance he has learnt to settle better and better.
"He has built up a good relationship with Tom [Queally] and they respect each other. Tom says 'go' to Frankel now, not the other way around.
"You dream about horses like Frankel. It is a wonderful to take him to York and I am confident that the Yorkshire crowd will come to see him."
Frankel is no bigger than 1-7 with William and bet365 to score at York, though Ladbrokes and Paddy Power offer 1-8.
