The Poule D'Essai Des Pouliches (the French 1,000 Guineas) runner-up was competing in her ninth race of the season but showed no signs of fatigue returned to the same course as her previous victory, the Group 3 Lanwades Stud Fillies Stakes late last month.
Last year's Blandford winner Manieree led into the straight and saw off the challenge of Chrysanthemum before Up and Caponata came through full of running.
The 3-1 joint favourites duelled right the way to the line but it was Up who battled to a desperately narrow victory.
Aidan O'Brien said: "She's a tough filly and we thought that a mile and a quarter would be as far as she would stay.
"Joseph was riding her to get the trip as we knew the last 50 yards were going to be a long way for her.
"The Breeders' Cup Filly and Mare Turf would suit her and she might not run before as she's been very busy lately."
Up has already run once in America, finishing sixth in the Grade 1 Beverley D. Stakes at Arlington last month.
